I think an Ayako route is highly unlikely, because it would require them to more-or-less entirely ignore Zero and all of the main characters from it in favour of a new character who isn't even mentioned there.
A Rider route is also unlikely, because of Rider's attachment to Sakura, and general reluctance to trust people. I can see her coming to like Shirou, but I find it hard to see her pursuing a romance with him when Sakura still needs him.
Well out of the three: HF would have been a terrible sequel because it has zero weight in terms of the ideal stuff (and a lot of other stuff) unless you've already gone through UBW, it's just too reliant on the previous routes to stand up on its own. Fate... I guess they just didn't want to retread the ground of the previous series. Also Fate's pretty short and the UBW movie was a joke in terms of adapting what that route was actually about.
Really the only way to have a proper sequel to Zero is Fate/Stay Night, not one of the routes, since Zero was designed to be a prequel to the whole thing. Having a UBW series and then a HF movie (1 or more, still don't know) seems like the best compromise they really could have done.
